Fixed this, kinda. I discovered there was an entry in the Communicator.adm GPO template to manually specify the address book location by pointing it directly to the server instead of the pool URL. Did the same with group expansion. Still doesn't explain why it doesn't work to begin with, but it is a nice functional work around.

If you get nothing - or an IIS error, on the Lync server, try opening the same URL. Open IIS on the Lync server, ensure App Pools are running. In IIS, also ensure the appropriate web sites are running. Check eventlog for errors/cert issues etc. This should give you some direction of where to look next.
